The performance of orally inhaled and nasal drug products (OINDPs) can be directly influenced by ambient temperature, humidity and electrostatics found in the laboratory, as highlighted by the US and European Pharmacopoeias. EnviroMate is an accurate, efficient, low maintenance solution that addresses these issues with considerable value for scientists faced with:
  • Variable lab conditions or inadequate climate control
  • OINDPs with high sensitivity to temperature, humidity and/or electrostatics, such as powders and aqueous formulations where hygroscopicity and evaporation can be an issue

Delivered dose uniformity (DDU) testing and aerodynamic particle size distribution (APSD) measurement by cascade impaction are critical tasks for the inhalation community. Variability in the test environment can affect the dose emission and aerosol generation performance of OINDPs, and APSD measurement methods, compromising the integrity of these tests. By tackling this source of variability EnviroMate reduces the risk of erroneous data and costly testing delays. EnviroMate is a compact, movable, benchtop unit with a spacious, clear-view chamber, large hinged front door, and slot-sealed rubber entry ports. It comfortably accommodates the Inhaler Testing Workstation™, all types of cascade impactor (including preseparator and mixing inlet as required) with dedicated side wall ports and quick-connectors for interfacing with externally located ancillaries. EnviroMate accurately controls and maintains uniform temperature (± 2°C) and humidity (± 5% RH) by circulating air through the entire chamber, whilst an integrated anti-static system helps to minimise electrostatic effects. The unit contains a self-regenerating desiccant dehumidifier and is refrigerant-free, easing maintenance and environmental impact. “Controlling environmental conditions precisely during the testing of OINDPs is crucial, but is typically complex and expensive,” says Mark Copley, CEO, Copley Scientific. “The EnviroMate is a pragmatic, well-engineered, highly effective option tailored specifically to inhaler testing requirements. The Taskforce is a collaboration of over 30 members including patients, health care professionals, the voluntary sector and professional associations all working together to represent the needs of people with lung disease. The Taskforce’s Industries Forum is a group of representatives from the pharmaceutical, diagnostics, devices, and digital industries that works alongside the Taskforce to improve the diagnosis, treatment, and care of patients living with lung disease in England. Company experts will join regular meetings of the Industries Forum to share relevant industry views, intelligence, and experience. Currently the subject of intense research activity, particularly for generics manufacturers, nasal products deliver drugs for both local and systemic action. The new range of accessories allows customers to:
  • Reproducibly measure the delivered dose uniformity (DDU) of nasal sprays, in line with regulatory requirements, using automated and manual methodologies.
  • Easily establish robust, automated nasal drug product test set-ups to measure the aerodynamic particle size distribution (APSD) of the respirable fraction of the dose.
  • Improve in vitro-in vivo correlations (IVIVCs) and study regional droplet deposition in the nasal cavity, to optimise drug delivery via a target pathway.
DDU measurement for nasal sprays calls for firing of the product into a vertically mounted sampling apparatus. The new Nasal Spray Dose Collector (NSDC) and Nasal Spray Waste Collector (NSWC) have been designed specifically for this purpose. While useful for manual DDU testing, they can be integrated with the Vertus II/Plus automated shake and fire system in a highly productive test set-up that maximises the reproducibility of DDU measurements while alleviating the health and safety risks associated with repeated manual nasal spray product operation. The new Alberta Idealised Nasal Inlet (AINI) is an important step forward in terms of achieving more clinically representative nasal product testing and is particularly valuable for the demonstration of bioequivalence. Building on the reputation of the Alberta Idealised Throat, the AINI was originally developed by Professor Finlay from the University of Alberta and has an idealised clinically validated geometry created from Computer Tomography (CT) scans of individual subjects. It is made up of four individual regions – the vestibule, turbinate, olfactory and nasopharynx – and mimics deposition behaviour in each while simultaneously generating a sample that accurately reflects the respirable dose. Manufactured from aluminium, the AINI has a robust, leak-free, easy-to-use design. It can be used with a filter, such as the Fine Fraction Collector, to quantitatively determine drug deposition by chemical recovery and assay or interfaced with a cascade impactor to representatively measure the APSD of the respirable fraction. Further accessories additionally enable interfacing of the Vertus II/Plus with any routinely used cascade impactor, via an expansion chamber or AINI, for representative measurement of the APSD of the sub-10 microns, respirable fraction of the dose. Such testing is essential to quantify the risk of drug delivery via the lung, rather than via the nasal cavity. As with DDU testing, use of the Vertus II/Plus with cascade impactors such as the Next Generation Impactor and Andersen Cascade Impactor for this application, reduces measurement variability, boosts productivity, and alleviates health and safety concerns. The Mixing Inlet allows application of a patient-representative inhalation profile during aerodynamic particle size distribution (APSD) measurement and is used to achieve better in vitro in vivo correlations (IVIVCs). The new, one-piece, all 316 stainless steel construction is designed to be more robust and chemically inert for the drug recovery process. The TPK™ 2100 Critical Flow Controller is a key accessory for flow rate control in dry powder inhaler testing (DPI) that now offers enhanced logging and recording capabilities as a result of the latest firmware update (Version 2.00). Cascade impactors are precision instruments that operate at a constant volumetric air flow rate. The Mixing Inlet decouples flow through the test product from that entering the impactor making it possible to apply a clinically realistic inhalation profile during measurement while meeting the constraint of constant flow, at a calibrated rate, at the inlet to the impactor. Available in both ACI and NGI versions, the new Mixing Inlet has a 316 stainless steel construction to aid drug recovery and optimise drug/solvent compatibility. A one-piece, rigid design offers enhanced induction port support and protects the sharp edge of the inner cone from damage; the avoidance of internal ‘O’ rings also eliminates a potential source of leaks. By gently mixing sample-laden air with any supplementary air required, the Mixing Inlet produces a homogeneous test flow for the impactor, while minimising internal drug losses. The key function of the TPK™ 2100 is to control the air flow entering the impactor for DPIs, ensuring stable, critical flow and a defined pressure drop across the inhaler - 4 kPa when applying compendial test methods. This pressure drop (P1) is now recorded and logged for every test providing a traceable record of compliance and correct inhaler operation. Cascade impaction is used to determine the aerodynamic particle size distribution (APSD) of all orally inhaled products; a critical quality attribute. The NGI Assistant automates the associated drug recovery process, making the conversion of NGI samples to solutions for HPLC analysis a simple, reproducible ‘load and go’ operation. The latest model offers shorter cycle times – boosting throughput and return on investment – and a simplified, easy-to-use interface. It frees analysts up for higher-value work while at the same time improving data integrity by eliminating costly errors and operator-to-operator variability. The NGI Assistant holds up to three complete NGI cup trays, but can also accommodate induction ports and preseparators, as required. Solvent is automatically dispensed by a robust, low maintenance liquid handling system to a volumetric accuracy of ~±1%. The proven gentle rocking platform ensures complete dissolution of the drug and aliquots of the resulting solutions are then aspirated into standard HPLC vials for direct analysis. The new system can accommodate dual HPLC vial racks, offering the capability to collect both primary and back-up samples for each NGI test undertaken. It is compatible with a wide range of solvents and specified with vials for industry-standard HPLC systems (Waters and/or Agilent); customisation for alternative vials is also available on request. Substantially upgraded Windows-based software provides an intuitive interface, making it easier to customise methods and safeguard data integrity via a range of security and electronic records management features.
