The new nasal dose uniformity sampling apparatus (DUSA) interface plate enables simple, leak-free connection between a standard DUSA and the nasal drug product to ensure complete dose capture. Designed for use with a Vertus II/Plus, Copley’s automated shake and fire system, it supports automated, high productivity, high repeatability testing. Introduction of the new interface means that all routine test set-ups for nasal drug products are now easily automated using Copley’s complete integrated solution. Dose uniformity is a critical quality attribute for nasal drug products because of its direct impact on clinical efficacy. Dose uniformity measurements involve vertical or near vertical firing of the nasal drug product and capture of the released dose. Leakage and loss of dose, back on to the product nozzle, is a common problem, as is variability arising from manual product actuation. The Vertus II/Plus is a well-established fully automated shake and fire system for nasal drug product actuation. Automated actuation of the nasal drug product reduces analytical variability, at the same time improving test productivity and reducing health and safety risks for analysts. With the introduction of the new interface, the Vertus II/Plus can now be used with:
  • A standard metered dose inhaler DUSA – as specified in recent revisions to the Ph. Eur. and USP, which are more prescriptive than preceding versions with respect to dose capture.
  • A Nasal Spray Dose Collector – an accessory designed specifically for nasal drug product testing that combines highly efficient drug capture and ease-of-use.
  • A Nasal Spray Waste Collector – for firing to waste under representative conditions.
Together these products represent a complete solution for dose uniformity for nasal drug product dose uniformity testing. Cascade impactors determine aerodynamic particle size distribution (APSD), a critical quality attribute for inhaled drug delivery. Inhalytix™ is flexible, user configurable and suitable for all cascade impactors in routine use, for both standard and bespoke test set-ups and methods. It handles the entry, analysis and reporting of APSD data to company and regulatory standards, in accordance with draft USP Chapter <1603>, Ph.Eur. Chapter 2.9.18 and relevant US FDA guidance. The software also serves as a database for laboratory cascade impactor inventory enhancing data traceability and integrity. Cascade impaction involves quantitative recovery and chemical analysis (typically by HPLC) of the size fractionated aerosol from an inhaled product to generate a drug specific APSD. The technique generates substantial quantities of data. Inhalytix™ is the successor to Copley’s highly regarded CITDAS™ software, built from the ground-up, drawing on the company’s established expertise in this area. It uses the same core calculation protocols but offers significantly enhanced functionality allowing customers to configure:
  • Reports – choosing which metrics to include to ensure the consistent and informative delivery of results, to meet regulatory requirements.
  • Equipment – selecting from a library of test equipment to handle standard and complex equipment configurations.
  • Methods – to combine detailed product information with reporting and equipment configurations, and to define metrics such as stage groupings and fine particle dose.
Interactive routine analysis, with a defined method, proceeds via a simple three-step process: Prepare, Execute and Analyse. Data can be entered manually or imported and comparison tools simplify assessment of the results, especially when comparing test and reference products in the case of generics. Using the equipment database users can link mensuration data, which quantifies changes in the critical dimensions of an in-use impactor, to specific, serialised impactors. This capability enhances data traceability and makes it possible to use actual rather than nominal dimensions to calculate APSDs, with greater accuracy. Inhalytix™ incorporates new metrics such as Impactor Sized Mass (ISM), Large Particle Mass (LPM), Small Particle Mass (SPM) and LPM/SPM ratio, amongst others and handles a broad array of impactors and components, including abbreviated impactors such as the Fast Screening Andersen (FSA). The new Nasal Spray Dose Collector (NSDC) and Nasal Spray Waste Collector (NSWC) have been designed specifically for this purpose. While useful for manual DDU testing, they can be integrated with the Vertus II/Plus automated shake and fire system in a highly productive test set-up that maximises the reproducibility of DDU measurements while alleviating the health and safety risks associated with repeated manual nasal spray product operation. The new Alberta Idealised Nasal Inlet (AINI) is an important step forward in terms of achieving more clinically representative nasal product testing and is particularly valuable for the demonstration of bioequivalence. Building on the reputation of the Alberta Idealised Throat, the AINI was originally developed by Professor Finlay from the University of Alberta and has an idealised clinically validated geometry created from Computer Tomography (CT) scans of individual subjects. It is made up of four individual regions – the vestibule, turbinate, olfactory and nasopharynx – and mimics deposition behaviour in each while simultaneously generating a sample that accurately reflects the respirable dose. Manufactured from aluminium, the AINI has a robust, leak-free, easy-to-use design. It can be used with a filter, such as the Fine Fraction Collector, to quantitatively determine drug deposition by chemical recovery and assay or interfaced with a cascade impactor to representatively measure the APSD of the respirable fraction. Further accessories additionally enable interfacing of the Vertus II/Plus with any routinely used cascade impactor, via an expansion chamber or AINI, for representative measurement of the APSD of the sub-10 microns, respirable fraction of the dose. Such testing is essential to quantify the risk of drug delivery via the lung, rather than via the nasal cavity. As with DDU testing, use of the Vertus II/Plus with cascade impactors such as the Next Generation Impactor and Andersen Cascade Impactor for this application, reduces measurement variability, boosts productivity, and alleviates health and safety concerns. Edward Brundrett, a new technical sales executive is working to enhance every aspect of the sales process while Mark Lo brings fresh advanced mechanical engineering knowledge to the design team. “We’re committed to providing a sales team that combines the technical expertise and responsiveness needed to serve our customers effectively and we are delighted to welcome Edward to it,” said Richard Postlethwaite, Chief Operating Officer, Copley Scientific. “As global leaders in testing equipment for inhalers and other dosage forms, we routinely break new ground with innovative products for faster, more reliable and more patient representative testing. Our knowledgeable sales team supports customers in identifying which products will help them to best address the challenges they face, and we are committed to investing in its growth and evolution.” With a reputation for continuous improvement, Copley has recently added market-leading automation expertise to its portfolio with the acquisition of Novi Systems.
